Effects of Antioxidants on the Viability of the Human Neuroblastoma SH-SY5Y Cell Culture under the Conditions of Heavy-Metal Toxicity

چکیده
Protective action of antioxidants (mexidol, carnosine, N-acetyl cysteine) and the metal chelator Ca, Na 2 -EDTA was studied in the culture of human neuroblastoma SH-SY5Y cells after the addition of salts of heavy metals—lead, cadmium, cobalt, and molybdenum—to the culture medium. Cells were incubated with heavy metals and protectors for 24 h, and cell viability and cell death were evaluated. All the metals lowered cell viability in a concentration-related manner. Different protective agents were studied based on this model. The most pronounced capability of increasing the cell viability in conditions of heavy-metal toxicity was demonstrated by N-acetyl cysteine (the protective effect was demonstrated at the concentrations 0.5-1.0 mM and higher). Protective potential of carnosine was somewhat lower and that of mexidol was minimal. Rheum turkestanicum Janisch Root Extract Mitigates 6-OHDA-Induced Neuronal Toxicity Against Human Neuroblastoma SH-SY5Y Cells
Background and Objective: Rheum turkestanicum (R. turkestanicum) has been known to reduce inflammation and has antioxidant properties such as protective effect in neurons. This study aimed to determine the effects of R. turkestanicum on neuronal toxicity induced by the pro-parkinsonian neurotoxin 6-hydroxydopamine (6-OHDA) in neuroblastoma SH-SY5Y cells. Materials and Methods: MTT and DNA frag...
متن کاملInvolvement of Mu Opioid Receptor Signaling in The Protective Effect of Opioid against 6-Hydroxydopamine-Induced SH-SY5Y Human Neuroblastoma Cells Apoptosis
Introduction: The neuroprotective role of opioid morphine against 6-hydroxydopamineinduced cell death has been demonstrated. However, the exact mechanism(s) underlying such neuroprotection, especially the role of subtype receptors, has not yet been fully clarified. Methods: Here, we investigated the effects of different opioid agonists on 6-OHDA-induced neurotoxicity in human neuroblastoma...
متن کاملP 36: Protective Effects of Crocin on D-Galactose Induced Aging Model in Human Neuroblastoma Cells
Introduction: D-galactose (D-gal) is well known as an appropriate agent to induced aging effects in the in vivo and in vitro models. In the present study, we selected crocin, the main constituent of Crocus sativus L. (Saffron), against D-gal cytotoxicity in human neuroblastoma SH-SY5Y cells. Materials and Methods: Cells were pretreated with crocin (25-500 µM) for 24 h and then expos...
متن کاملThe neuroprotective effect of BSA-based nanocurcumin against 6-OHDA-induced cell death in SH-SY5Y cells
Objective: Parkinson’s disease (PD) is regarded as the second most common neurodegenerative disease affecting elderly population. There is a tendency toward finding natural cures to suppress the initiation and progression of this disease. Some epidemiological studies indicated lower incidence of PD in populations that consume curry. Curcumin, as the main ingredient of turmeric, has been suppose...
متن کاملGlycogen synthase kinase-3β may contribute to neuroprotective effects of Sargassum oligocystum against amyloid-beta in neuronal SH-SY5Y cells
Glycogen synthase kinase (GSK)-3β mediates amyloid-beta (Aβ) and oxidative stress-induced neurotoxicity in neurodegenerative disorders. Natural products with antioxidant activity, such as Sargassum (S.) oligocystum may modulate GSK-3β enzyme and protect against Aβ-induced neurotoxicity. Therefore, we aimed to assess the neuroprotective effects of a methanolic extract of S. oligocystum against A...
